Schaudinn 's fluid Formula Mercuric chloride, saturated aqueous solution Absolute alcohol 2 parts 1 part This fixative has been popular for many years as a cytoplasmic fixative for wet smears. It is not recommended for tissue, being harsh in action and causing excessive shrinkage ; probably due to the time of exposure needed for penetration. Tissue or smears need treatment with iodine-alcohol and sodium thiosulphate to remove mercury deposit (page 33). Wet smears are well fixed in 10-20 minutes and unless too thick they rarely become subsequently detached from the slide.

It is a protein precipitant, forming picrates with amino acids, including those in nucleoproteins. Although it causes shrinkage of tissues, it enhances cytoplasmic constituents, giving especially brilliant results with trichrome stains. Picric acid also dyes tissue yellow, sometimes of benefit when handling small fragments. The yellow colour is easily removed from sections. Picric acid is often employed in compound fixatives to enhance the preservation of glycogen, but may render RNA resistant to ribonuclease digestion.

Both methacrylates and epoxy resins have been used. Resin embedding is also used for embedding very hard tissues, such as undecalcified bone and teeth which are to be cut with a diamond impregnated disc and ground to a suitable thickness, or cut on a specially designed microtome at 10 μπι. Gelatin Fragments of friable tissue to be cut on the freezing microtome or cryostat may be first embedded in gelatin. Gough and Wentworth (1949; 1960) developed a method of producing sections from whole organs, such as lung, embedded in gelatin.
